172DEVELOPMENT AND CHARACTERIZATION OF CHLORAMBUCILLOADED GELATIN NANOPARTICLES FOR TARGETED DRUG DELIVERYChlorambucil-loaded gelatin nanoparticles (Chlor-GNPs) were developed and characterized in this study in order to enhance therapeutic efficacy and minimize systemic toxicity. Chlorambucil, a chemotherapeutic agent used in treating chronic lymphocytic leukemia and other cancers, suffers from poor solubility and significant side effects due to nonspecific distribution. The use of gelatin nanoparticles as a delivery vehicle was investigated to address these limitations. Gelatin nanoparticles were prepared using the desolvation technique, with Chlorambucil incorporated during the nanoparticle formation process. The formulation parameters, including gelatin concentration, pH, and crosslinking agent concentration, were optimized to achieve maximum drug loading efficiency and desired particle size. The optimized Chlor-GNPs were characterized for particle size, polydispersity index (PDI), zeta potential and drug release profile. Cytotoxicity assays on cancer cell lines showed that Chlor-GNPs significantly enhanced the anticancer activity of Chlorambucil compared to the free drug, with improved cell uptake and apoptosis induction.
